I'm using Ulead Video Studio 9 and I can't seem to save an AVI file to WMV format.
I can select Video Save Options and then select Profile - Customize and the Profile Manager allows me to create my desired Profile.
However, this Profile does not subsequently appear in the Profile dropdown box.

no, it won't save your settings that way.
You need to use "Tools", "Make Movie Manager" and create a new profile by clicking the "New" button in the "Make movie Templates Manager" dialogue box that appears. Input all your settings, and when you close the dialogue box your new profile will be saved.
Then, when you use "Share", "Create Video File" your new profile will appear at the bottom of the list, just above "Custom".

There are some conditions under which you can create a profile that is either incompatible or just plain doesn't get enumerated by UVS.
For instance, if you uncheck the Audio checkbox while editing the profile in order to create a video-only WMV file, UVS will not present this profile as a choice (the workaround is to choose an audio format of 0 Kbps Mono). In UVS 8, you also could not specify a framerate and had to use "Same as input file", or the profile would not appear.
