Create new layer. The tab is on the bottom of the layers list. Drag it below your image. Use the flood fill (bucket) tool. The color it will use is the top color square on right click. The bottom on left click.
If the image want allow you to put it above the new layer, right click on the layer at the layer list and select promote background layer. Though if the image has a transparent background it's format may be png so this step would more then likely not be necessary.
Be sure to save it using a format that will retain the layers psd, psp image. This is so you can edit the layers at a latter date if you choose. If you save a png, targa, jpg all layers will be lost making the image very difficult to edit.

What kind of layer (rater, vector, etc.) do I create?
When you say drag it below your image - is that on the bottom of the layers list?
