I don't really know what these do so should they all be ticked? and what about the Performance Optimization?

The main reason these are optional is that they don't work all the time for all video sources and projects. Best is to try them; when they work, rendering/sharing is faster (that's all they do).

Yes, you can turn it all on. It uses DXVA2 Acceleration, and will automatically disable what isn't supported. You can search the configuration files and see what it actually turned on. On my i7/GTX machine, barely anything was actually enabled, so I don't think it makes a huge difference... at least not for timeline/NLE playback performance.
