If it helps - I've managed to achieve the similar effect (different subject matter), but only as a two-stage process, as follows:

1. Film hands
2. Use paining creator to draw motion of "pen" in a colour of your choosing
3. Place painting creator object in overlay, hands in main video channel
4. Render to video file of your choosing.

THEN

5. Start a new project
6. Place photos/video in main video channel
7. Place Hands+pen rendered file in over channel
8. Set Chroma-key colour to pen colour

and Bob's yer Auntie :D

I can't help but feel there should be a slightly slicker way of doing this, but it does seem to work pretty well.

Well! I have had a go at this. Didn't have time to do the hands but got the main part about right I think. When I do the hands I shall have a go at filming them with a one color background and using Chroma Key.
My effort is here, needs a bit of practice painting smoothly with a mouse. Not done that much before:
