Ok, sorry, do you have any DLA (Drive Letter Access) software installed? Off the top of my head I can think of Nero InCd or Sonic...something or other.
These progams allow you to "drag'n'drop" files to the CD/DVD burner on the fly. They have a habit of seizing control of the drive at start up and being very reluctant to let other programs get access to the drive.
The Sonic one can be disable from the system tray but InCd hasto be uninstalled.

Apart from Lance's suggestion, have you tried going into Control Panel > System > Hardware > Device Manager and uninstalling the driver for the burner. Then rebooting and letting Windows find your drive and reinstall the drivers? Or turning off your computer, opening up your computer case and pulling the cables from the back of the drive. Then rebooting, and turning off the computer again, then reconnecting the drive cables and rebooting so that Windows will similarly go through the process of recognising the drive as new hardware?
